Inhibitors of Apoptosis Proteins (IAPs): Clinical Significance in Cancer Treatment Research
نویسندگان
چکیده
Apoptosis is a process, which involves a sequence of cellular changes, which ultimately lead to cell death. This programmed cell death is a normal phenomenon required for growth of an organism. Inhibition of apoptosis can result in a number of cancers, inflammatory and autoimmune diseases and viral infections. Inhibitors of apoptosis proteins (IAPs) are a family of structurally and functionally related proteins, which play a crucial role in apoptosis (programmed cell death), proliferation and angiogenesis. Till date 8 IAPs have been identified (Survivin, XIAP, Livin, cellular IAP 1 and 2, ILP-2, NAIP and BRUCE/Apollon). The current review discusses individual protein in details with respect to its structural features, functions and clinical significance. These proteins; especially survivin, XIAP and Livin have been found to express in wide range of malignancies and hence taken as a target of interest by various research groups. The review also highlights the various Phase1 and 2 studies of new therapeutic agents that are being developed either as a monotherapy or in combination with existent drugs, which target these IAPs.

Inhibitor of apoptosis proteins: translating basic knowledge into clinical practice.
The inhibitor of apoptosis proteins (IAPs) are a family of antiapoptotic proteins that bind and inhibit caspases 3, 7, and/or 9, but not caspase 8. Growing evidence also indicates that IAPs also modulate cell division, cell cycle progression, and signal transduction pathways. As our basic understanding of IAPs has increased, the knowledge is being translated into clinically useful applications ...
متن کاملSuppressive Effect of Constructed shRNAs against Apollon Induces Apoptosis and Growth Inhibition in the HeLa Cell Line
Background: Cervical cancer is the second most common female cancer worldwide. Inhibitors of apoptosis proteins (IAPs) block apoptosis; therefore, therapeutic strategies targeting IAPs have attracted the interest of researchers in recent years. Apollon, a member of IAPs, inhibits apoptosis and cell death. RNA interference is a pathway in which small interfering RNA (siRNA) or shRNA (short hairp...
متن کاملThe effect of podophyllotoxin as an inducer of apoptosis using molecular docking method
Background and aim: Podophyllotoxin is used as one of the main treatments for genital warts. It is a precursor of etoposide and teniposide, which is used in the treatment of various cancers. Despite a large number of cancer studies, the exact mechanism of podophyllotoxin remains unknown. Chemotherapy drugs reduce cancer cells by inducing apoptosis. The regulation of the apoptotic pathway has be...
متن کاملHDAC Inhibitors and Heat Shock Proteins (Hsps)
Epigenetic alterations, including DNA acetylation, hypermethylation and hypomethylation, and the associated transcriptional changes of the affected genes are central to the evolution and progression of various human cancers, including pancreatic cancer. Cancer-associated epigenetic alterations are attractive therapeutic targets because such epigenetic alterations, unlike genetic changes, are po...
متن کاملNovel smac mimetic APG-1387 elicits ovarian cancer cell killing through TNF-alpha, Ripoptosome and autophagy mediated cell death pathway
BACKGROUND Ovarian cancer is a deadly disease. Inhibitors of apoptosis proteins (IAPs) are key regulators of apoptosis and are frequently dysregulated in ovarian cancer. Overexpression of IAPs proteins has been correlated with tumorigenesis, treatment resistance and poor prognosis. Reinstalling functional cell death machinery by pharmacological inhibition of IAPs proteins may represent an attra...
